微波辐射法合成乙二胺四乙酸-α, δ-(2-甲基-5-硝基咪唑-1-乙醇)二酯

Synthesis of Ethylenediamine-Tetra-Acetate Diester of α, δ-(2-Methyl-5-Nitroimidazole-1-ethanol) by Microwave Radiation Method

摘要: 以乙二胺四乙酸双酐(EDTAA)和2-甲基-5-硝基咪唑-1-乙醇(甲硝唑, MN)为原料,采用微波辐射法合成乙二胺四乙酸-α, δ-(2-甲基-5-硝基咪唑-1-乙醇)二酯(EDTA-2MN).设计正交实验,探讨了物料比、反应时间和反应温度等对收率的影响.结果表明,在EDTAA和MN物质的量之比为1∶2.5、反应时间30 min、反应温度110 ℃时,收率达到80%以上.与常规的酯化反应相比,该方法具有反应时间短、收率高和重复性好的优点.并采用红外光谱、核磁共振氢谱、质谱及元素分析对产物进行了表征.

Abstract: In this paper, ethylenediamine-tetra-acetate diester of α, δ-(2-methyl-5-nitroimidazole-1-ethanol ) (EDTA-2MN) is synthesized from ethylenediamine-tetra-acetic acid anhydride (EDTAA) and 2-methyl-5-nitroimidazole-1-ethanol (metronidazole,MN) by microwave irradiation method. Based on the designed orthogonal experiment, the effects of the molar ratio of the reactants, reaction time and reaction temperature on the yield are investigated. The results indicate that the yield is more than 80% under the reaction condition as follows: the molar ratio of EDTAA to MN is 1∶2.5, the reaction time is 30 minutes and the reaction temperature is 110 ℃. Compared with the conventional method of esterification reaction, the distinct advantages of the microwave irradiation method are short reaction time, high yield and good repeatability. And the structure of the product is characterized by FT-IR, 1HNMR, MS and elemental analysis.
